Australia had done well to take the game through to day five but the partnership of Moises Henriques and Nathan Lyon could add only nine more runs before Ravindra Jadeja dismissed Lyon.
Although Virender Sehwag and Murali Vijay departed cheaply, India eased to victory with Tendulkar batting like a man back in form and in complete control alongside Cheteshwar Pujara.
The second Test in the series gets underway in Hyderabad on Saturday.
